Probing GLP1R signaling pathways with unusual ligands

As an established target for the treatment of type 2 diabetes mellitus (T2DM), the Glucagon-like peptide-1 receptor (GLP1R) has attracted substantial interest. GLP1R is a seven-transmembrane G-protein coupled Receptor (GPCR) that activates multiple signaling pathways including β-arrestin recruitment and Gαs signaling. Using non canonical techniques (and combinations thereof) such as dimerization, chimera generation, side-chain stapling, and backbone modifications, I generate peptidic ligands targeting the GLP1 Receptor to probe its function.
