Determining Cellular Functions of Diphthamide Using a Proteomics Approach

Diphthamide is a conserved post-translational modification currently only known to be found on one protein in the cell and whose biological function remains poorly understood. Diphthamide and its biosynthetic enzymes have been implicated in a number of different cellular pathways; however, there has been no systematic, cell-wide search for diphthamide modification on other proteins. We are taking a functional proteomics approach to elucidate other proteins with diphthamide modification and cellular pathways involving diphthamide biosynthetic enzymes in order to better understand the biological function of diphthamide.
